Since the hydrophobic characteristics of the target product are difficult to obtain directly, when designing a hydrophobic chromatography process, pre-experiments should be carried out with fillers containing different hydrophobic ligands. The hydrophobicity of the hydrophobic ligand butyl-octyl-phenyl increases sequentially, and the hydrophobicity of the target product can be predicted through the combined adsorption test with different hydrophobic fillers. After understanding the hydrophobicity of the target product, pretreatment of the target product should be carried out. Generally speaking, the target product will be combined with a hydrophobic filler in a high-salt system and eluted under a low-salt condition. Therefore, it is necessary to optimize the salt concentration during sample loading(ammonium sulfate is usually selected as the pretreatment salt for sample loading). In addition, after preliminarily determining the type of ligand and the salt concentration required for sample loading, the gradient elution from high salt to low salt can be used for filler screening and process optimization. The follow-up can be further optimized through experimental design.
